This technology achieves an early diagnosis of hepatocellular carcinoma by comprehensively sequencing and analyzing abnormally hypermethylated CpG islands in the peripheral blood free DNA of patients, and is a breakthrough in cancer diagnosis methods. The results were published online in the October 30 issue of Cell Research and will be officially published in the form of a journal cover article.

At present, the sensitivity of DNA methylation detection technology is low, but it is far from meeting the requirements for high-throughput detection. The MCTA-Seq technology cleverly breaks through this bottleneck. By selectively amplifying the methylated CpG short tandem CGCGCGG sequence and subsequent high-throughput sequencing analysis, MCTA-Seq can simultaneously detect nearly nine thousand CpG islands in one reaction with a detection limit as low as 1 or 2 Genomic DNA of cells. In addition, because MCTA-Seq's three-step library-building reaction can be completed in a PCR tube and requires only shallow sequencing, it is a simple, economical and broad application technology for free DNA sequencing.

Hepatocellular carcinoma is one of the highest incidence of cancer in the world, with the third highest mortality rate. Due to the large number of people infected with chronic hepatitis B virus, the incidence of hepatocellular carcinoma has been high in China. At present, serum alpha-fetoprotein (AFP) is the most commonly used early screening marker for hepatocellular carcinoma in the clinic, but the positive rate is only 60-70%. Therefore, it is necessary to develop new hepatocellular carcinoma early screening biomarkers.

By combining two types of plasma CpG island markers, the sensitivity of the MCTA-Seq technique for the diagnosis of hepatocellular carcinoma was 94% with a specificity of 89%. More importantly, MCTA-Seq successfully performed a correct diagnosis of all 15 patients with hepatocellular carcinoma with false negative AFP in this study, and was able to identify the tissue source of the tumor by analyzing its CpG island methylation pattern.
