As an effective gene editing method, CRISPR-Cas9 has great potential in disease prevention and treatment, but its off-target effect must be considered in clinical transformation. Traditional biological methods such as electrophoresis or sequencing can be used to study the targeting of Cas9, but the results are mostly static and average. The latest article published in the March 3019 issue of the journal Nature Structural and Molecular Biology, using the latest fluorescent aperture technology, using a light-operated DNA template, combined with a confocal fluorescence microscopy imaging system, enables dynamic observation at the single molecule level.
